Closing a vent will save a little money but there are risks. Heating and cooling systems need a certain amount of air flow. If too much air is restricted, whether it be by having a dirty filter, closing a vent or other, the system will not work right and it could cause long term damage. Each system is different but typically closing one vent is OK, two may be OK but more than that and you are increasing your chances of having a problem.
my husband swears i freeze him so he closes the vent in the bathroom. LOL
It depends on how much back pressure it creates. One school of thought you are not cooling another room so that will save some energy.
On the other hand you are making the blower motor work harder so it will use more current.
Assuming this room you are not cooling has one exterior wall you will not save much since the interior walls have no insulation.

I use a 2nd floor room for storage and I closed the a/c vent because it's not used as a living space. But in addition to closing the vent, I also kept the door closed. Real bad idea!
I went in there this past weekend and the room was very warm and had a clammy and damp feel, with an unpleasant odor to boot!
If any money is saved by closing the vent, I'm sure other unpleasantness creeps into the equation to cancel the savings out.
