If you want to find out if the cold water taps in your house are supplied direct off the rising main just turn off the main stop tap, if no water comes out of the taps then they are supplied by the rising main, if water comes out they are supplied via the cold water storage tank which as RichB says makes it unsuitable to drink.
It depends on how the plumbing in your house is set up. In many houses - especially older ones - cold water in the kitchen will be direct from the water main, but cold water in the bathroom will first pass through a storage tank in the roof, making it potentially unsuitable for drinking.

If the sink is clean and your not dipping it from the toilet, its the same water line.
In older properties lead piping was used upstairs to save on the cost of copper piping which is expensive. As we have outlawed lead this is no longer a problem. We brush our teeth in the bathroom so I cant see any problem with drinking it now.
