I guess what your asking is what size to build? I don't know your habits, if your a home body or if you are busy and just get home to sleep. House style? What to spend? Tornado alley?
I like a big big bathroom and kitchen, the most important for me. Living room and bedrooms need to be ample but closet size is so important in a bedroom. All things aside, and to make a thing small enough to keep clean easy, 25 feet by 40 feet. If I did it for myself I would do a single floor thich insulated concrete building with a flat of terrecotta roof, made to look like it is adobe. 8 inch thick minimum walls insulated will stay warm in the winter and cool in the summer and if done right will be safe beyond anything thrown at you from a tornado to a burgler. The proper plan would allow you to be "Green", and to spend next to nothing in heating and cooling, to be safe, and not cost any more than any other house it's size.

The question's too broad to answer off the bat:
Do you like cleaning? (Do you want to do it *all day*?)
Will there always be only one person living there, or might that change?
Do you like several levels, a basement, etc or do you want it all on one level?

One acre is a HUGE area for a house, even if you've got 23 pets and 14 kids. We've got 3 of us in a small house (98 sq metres = 1055 sq feet = about 1/40th of an acre) and that seems all right if the space is used well, but more would be easier with another baby on the way. Double that would be a biggish house for one person, but still manageable, and you'd have a decent sized yard.

You could, of course, subdivide the land and get 4 or more town houses on it; then there'd be a source of income as well (though I imagine it's out in the country so maybe not as potentially lucrative as a large city block...)
