B.
I would be a bit leary about the MDF. Maybe check out www.hgtv.com and see if there is any info from the woodworking guys?
It sure can. I did this when I built 2 bookcases last summer.
Someone suggested gorilla glue earlier -- I would stay away from that for bisquit applications. Bisquit joints get their strength when the bisquit swells as it absorbes the liquid from the glue. Gorilla glue expands to fill in the joint, but the bisquit itself will not absorb much and thus won't swell and your joint may remain loose.
Tests show (look in the most recent copy of Fine Woodworking) that yellow glues like Titebond II or Elmer's Woodworking glue produce stronger joints than Gorilla Glue.
I personally used Titebond II on the bookcases I built and it worked great.
Why bother? Titebond III will bond the two together so well that if you later attempt to break it off, the wood will split before the glued seam gives way (I know; I've done it.)
