How do you know if your basement is stubbed for a bathroom?

If it is plumbed for a bathroom, there will be a toilet flange on the floor. It's a 7" diameter white plastic disc looking object. Here's an example:

there should be pipes coming up through the cement that are capped...and all within a close range from eachother. a 3 inch for the toilet and a smaller one for the sink. and maybe another if it is prepped for a shower
look for a white plastic or black sticking out of the floor about 4 inches.
What you are asking is, is there a drain in the floor for the toilet and sink. They don't often put them in basements unless asked when being built. I had a bilevel home and we were plumbed for the bottom half. Even tho it was concrete blocks we finished with drywall etc. So, you'll have to ask a plumber how to put them in. Probably an upflow instead of downflow.
most times the basement is not plumbed you usually have to do that afterwards
