No. Plexiglass is just a grade of plastic. Most drill bits can drill through mild steel.
Just use a sharp general purpose drill bit (not a masonary, wood or auger type) and don't press too hard. Go nice and easy and it should make a nice clean hole.
If in doubt ask the hardware store people. They will give you what you need.:)

although plexiglass can be drilled with a 'normal' high speed steel drill bit, quite successfully, drill bit manufacturers do offer specialist drills.
To be honest, I can no longer remember whether the drill point was a 'flat' angle or a very acute one, (I think it was the acute angle) but the point angle was designed to avoid 'break through' splintering which can be a problem, especially in cold weather.
If you contact any industrial tool supplier (if you live in the UK then Cromwell Tools would be a good bet, they have numerous branches) and ask them they will advise the best type of drill bit to use.
We used to make waterproof switches in clear plexiglass and accuracy was paramount. If you just need the holes for a non-critical reason, then quite possibly a high speed twist drill of the correct size may well be good enough.

Note...always try to maintain a relatively warm atmosphere when drilling plexiglass...it can splinter or even shatter when cold.

if you drilling a small hole 3/8 then used a center point bit
for larger hole used a hole saw work great
