most codes call for a 1/4 in drop per running foot of pipe regardless of the diamiter.so a 4ft pipe would have a 1in drop.

The friction coefficent of the pipe along with the water pressure and velocity at the start of the pipe.

at a guess I would say 1 or 2 degrees should be fine.

But of course if also depends on how fast you want the water flowing in the pipe
Make it easy on yourself and use a level that has two lines on each side of the bubble, when you put the level on the pipe you need the edge of the bubble to touch the outside line and you will have the proper drain used in the plumbing trade. Good Luck
