I think you do, otherwise you're going to get dirt going between the tiles. This will encourage the growth of mold and give plants and weeds somewhere to take root. If it's wall tiles I would still say yes to keep them waterproof.
Yes. Whether they're floor or wall there needs to be a filler to stop water penetration under the tile.
