Do cotton garments shrink in the dryer?

Yes, even if they are preshrunk, they do shrink a little more.
The best way to dry them would be to smooth out most of the wrinkles and hang them up, Preferably outside on a windy day. (not so windy they blow away though)

Yes, they will shrink. Cotton expands a lot when wet. Then, when you remove that moisture, the cotton retracts greatly as well and causes shrinkage. The good thing is that cotton has excellent elongation. With normal wear, cotton items stretch a little. Something that comes out of the dryer feeling like it shrunk a little, usually feels fine once you wear it for a few minutes.
